I would like a Vampire Hunter D movie. I love the anime and the books seem very interesting.

Agree with Half-Life somewhat, although a movie of an already massively cinematic game may just cheapen the original experience.
Also, what if I pick like a book or a video game that wouldn't work as live action but would as an animated series or feature? Fuck it, I'm including those.

Monkey Island could work either as a finite movie series that tries to translate the games (which wouldn't be so great IMO) or as an ongoing TV show. Animated preferable to live-action because of voice-casting continuities and inevitable comparisons to PotC.
Wouldn't mind seeing on-screen translations of Y: The Last Man (in the pipeline still?), Transmetropolitan, Preacher, and 100 Bullets. The last one would probably work really well as a TV show due to the way its constructed. In fact, all of those would probably work better as mini-series rather than stand-alone movies or trilogies or whatever.

Have to agree with these. The Boys would be great if they could get away with the inevitable NC-17 rating. Waaay too much drugs and buggery not to get anything below an R rating.
Monkey Island as an ongoing animated series, good lord yes.
Y: The last Man, 100 Bullets and Preacher as finite tv shows would be excellent. Transmetropolitan has to be a couple of films I think (then again I'm only up to issue 47).
