with distortion the signal is (probably) compressed as it goes in, and boosted up to the point where it will always distort no matter how hard you play.
so gain would be good for van halen type stuff, and distortion for like nirvana or whatever.

Gain makes the wave bigger.
Distortion changes the wave shape.
Gain can cause the signal's shape to change, but only when you make the wave so big that the peaks start to clip.
Distortion can be applied at any volume level.
