Is this happening with ALL video files or just some?

Are you running Google Desktop by any chance?

I had this problem a while back and Google Desktop is always scanning the drives for new additions and changes. When it does this, it effectively "opens" the files (for want of a better word) which then stops a second program from opening the file as well.

Try turning off Google Desktop and trying again.

Again, as Xero said, it can also be a codec conflict. If you have G-Spot, run the file through this and it will tell you if you have the right codec.
